RSD = \%CV = \frac{s}{|\bar{x}|} \times 100\%, \quad s = \sqrt{\frac{\sum (x_i - \bar{x})^2}{n - 1}}sx̄nSEM“An analytical chemist measures 6 replicate dissolution assay samples: [99.8, 100.2, 99.5, 100.1, 100.4, 100.0] mg.”
Calculate central tendency and squared deviations with n-1 degrees of freedom.
Standard deviation represents absolute scatter around the mean.
RSD < 1.0% earns the highest analytical precision grade.
An RSD (%CV) of 0.31% easily satisfies the strict USP/ICH chromatographic assay criterion of ≤ 1.0% (Ultra-High Precision).
| Input Scale | Standard Output Range | Variance Band | Recommendation |
|---|---|---|---|
| Low Baseline | 0.00 – 25.00 | ±1.5% | Optimal for conservative estimations |
| Median / Standard | 25.00 – 100.00 | ±0.8% | Benchmark standard for commercial workflows |
| Elevated / High | 100.00 – 500.00 | ±0.4% | Recommended for high-throughput modeling |
| Extreme Maximum | > 500.00 | ±0.1% | Verify input constraints for boundary saturation |

Standard deviation depends on measurement scale and units. RSD expresses variability as a percentage of the mean, allowing direct comparison of precision across experiments with vastly different scales (e.g. microgram vs kilogram quantities).
According to USP and ICH guidelines, chromatographic assay repeatability typically requires RSD ≤ 1.0% or ≤ 2.0%. Biological assays and dissolution tests generally allow RSD up to 5.0%.
If the mean is zero, RSD is mathematically undefined due to division by zero. If the mean is negative, the absolute value of the mean is used in the denominator (|x̄|) so that RSD remains a positive percentage.
